Called pTCiMRFaΔCH in the article. In this misexpression plasmid, MRFa (small transcript of MRF) carrying a deletion is cloned upstream of 3.3kb brachyury regulatory element, to target notochord-specific overexpression. The deletion targets the Ci-MRF sequences encoding amino acids 365–385 (HYHH·····CKAC) (this deletion corresponds to bases 1093–1155 beginning with the ATG start codon of CiMRFb).
