Called pTCiMRFbNN in the article. In this misexpression plasmid, full lenght mutated MRFb (large transcript of MRF) is cloned upstream of 3.3kb brachyury regulatory element, to target notochord-specific overexpression. The mutation targets the sequence encoding the alanine398-threonine399 dipeptide encoded by Ci-MRF, replaced by an asparagine dipeptide. Original comment: This particular mutation was chosen because an asparagine dipeptide occurs at the corresponding position of the basic domain of related, but non-myogenic b-hlh proteins such as E12 and E47.
