Oocytes of a single individual of Halocynthia roretzi (Aomori, Japan) were fertilized by sperms of an another single individual and development was followed microscopically. Embryos were snapfrozen in liquid nitrogen at the appropriate stage and kept at -80°C. Total RNA was prepared with the AGPC method and the quality was checked with the Bioanalyzer, most samples had a RIN value of higher than 9. Strand specific libraries were constructed and the sequencing (50PE) was performed with Illumina HiSeq 2000 technology (BGI, Hong Kong, China).

Vegetal view of a Halocynthia roretzi 110-cell stage embryo probed for Mesp expression, injected with morpholinos against Pem and treated with GSK126, a selective inhibitor of EZH2 used to inhibit H3K27me3.
Mesp endogenous expression is lost in 46.2% of embryos without Pem and H3K27me3.
Original comment: Pem- and H3K27me3-deficient embryos showed no ectopic Mesp expression in germline cells (0%, n ​= ​13; yellow arrows).

Vegetal view of a Halocynthia roretzi 110-cell stage embryo probed for Brachyury expression, injected with morpholinos against Pem and treated with GSK126, a selective inhibitor of EZH2 used to inhibit H3K27me3.
Brachyury is expressed in B8.6, B8.5, A8.5, A8.6, A8.13 and A8.14 blastomer pairs, as in WT conditions, and ectopically in B7.5, B7.2 and B7.7.
Original comment: Embryos lacking Pem and H3K27me3 ectopically expressed Brachyury in B7.5 and other posterior vegetal cells, but not in germline cells (0%, n ​= ​8; yellow arrows).
The numbers represent the proportion of embryos showing ectopic gene expression in the germline.
