The J0.2 enhancer containing a mutated Onecut binding site was cloned upstream of the human β-globin basal promoter and LacZ reporter gene. This reporter construct is active in the central nervous system of Ciona robusta tailbuds and larvae. Only 29% of the embryos showed activity (65% for the WT enhancer).
