This construct tests the effect of the putative repressor sequence of Wnntun5 enhancer on FoxD.b enhancer. Four repeats of Wnttun5 repressor were inserted into the enhancer of FoxD.b, and the chimeric sequence was ligated to the Brachyury basal promoter upstream of GFP reporter gene. The insertion of the four repeats is directly downstream of the Tcf7 binding site cluster, as it is located in WT Wnttun5 enhancer.
