This construct was made to study the background activity of the basal promoter of Forkhead, widely used upstrean of LacZ reporter gene in pCES vector (pCES contains a multiple cloning site upstream of a forkhead promoter coupled to a LacZ reporter gene.). Original comment : This ‘background’ expression is inherent to the forkhead promoter of pCES and has been well characterised as LacZ expression in the head/neck mesenchyme and tail muscle cells , with some animals also showing LacZ in the centre of the sensory vesicle. This background becomes much weaker or is completely abolished when a regulatory element is driving the promoter.
